Het opkomend nationalisme aan het begin de negentiende eeuw vormde de aanleiding tot de oprichting van gedenktekens voor belangrijke personen uit de nationale geschiedenis. In 1852 onthulde Koning Willem III op het plein een door Louis Royer ontworpen gietijzeren standbeeld van Rembrandt. Het is het oudste nog bestaande standbeeld in Amsterdam en werd gegoten in één stuk, een bijzondere verrichting voor die tijd. In 1876 werd de naam Botermarkt gewijzigd in Rembrandtplein. Het beeld van Rembrandt heeft door de tijd heen op diverse plekken op het plein gestaan.
De van oorsprong Vlaamse beeldhouwer Louis Royer (1793-1868) maakte portretten van beroemde Nederlanders. Hij was medeoprichter van de Amsterdamse kunstenaarsvereniging Arti en Amicitiae in 1839.

Het oudste nog bestaande standbeeld in Amsterdam is ‘Rembrandt‘ op het Rembrandtplein, dat in 1852 is gerealiseerd. Kort daarop volgde het standbeeld voor Vondel in het Vondelpark. In de 19e eeuw werden in Nederland ongeveer 35 standbeelden en monumenten opgericht. In bijna alle gevallen gebeurde dit op particulier initiatief, de overheid speelde er nagenoeg geen rol bij. En er waren nauwelijks Nederlandse beeldhouwers die in staat waren dergelijke opdrachten naar tevredenheid uit te voeren, het is tekenend dat de uit Mechelen afkomstige beeldhouwer Louis Royer (1793 – 1868) maar liefst vijf standbeelden voor Nederland heeft ontworpen waarvan twee in Amsterdam: Rembrandt en Vondel (andere beelden: Michiel de Ruyter, Willem van Oranje en Laurens Janszoon Coster).
De periode van idee tot onthulling besloeg twaalf jaar. In 1841 werd het plan tot eerbetoon aan Rembrandt opgevat en een manifest zag het licht met als program het oprichten van het beeld. Beeldhouwer Louis Royer verwierf de opdracht, hij woonde destijds op de Oudezijds Achterburgwal en daar ontstond Rembrandt.
Huisvriend Jozef Alberdingk Thijm schreef in 1843 'Hij werkt van ’s morgens zessen tot ’s avonds negenen en vervalt zichtbaar, maar het werk stijgt ten top; het kind groeit ten koste van de moeder.' Een jaar later, toen het model klaar was, dichtte de huisvriend:
Ja God ! ja, dat is grootsch ! Dat is het schepsel waardig Eens door U-zelf gevormd. Sterk, vruchtbaar, edelaardig
De uitvoering verliep niet zonder problemen. Geld was bijeengebracht en voor zolang belegd in staatsobligaties. Maar de waarde daarvan kelderde zodanig dat overwogen werd het beeld in beton te laten uitvoeren. Gelukkig bleek het bedrag toch nog net toereikend om het in ijzer te laten gieten. De eerste gieting mislukte, er vielen zelfs gewonden, maar een tweede gieting in 1851 slaagde. Het beeld werd uit één stuk gegoten, voor die tijd een knap stuk werk. De naam van de meestergieter Ivar Jacobsen kwam daarom ook op de sokkel. Toen Rembrandt in 1924 een nieuwe, lagere sokkel kreeg, is Jacobsen van het voetstuk gevallen.
et sein voor de onthulling (‘ontblooting’) op 27 mei 1852 werd gegeven door koning Willem III nadat hij 'Leve het vaderland, hoezee' had geroepen. ’s Avonds was er een groot feest waar ruim 200 kunstwerken werden verloot, afgestaan ter financiering van het beeld. Ook was er vuurwerk voor het volk. Jan Schenkman (onderwijzer / illustrator) dichtte het volgende verslag: ‘Er waren Rembrandt-broodjes, Rembrandt-moppen, Rembrandt-taarten, Rembrandt-poppen, ’t zij van suiker, gips of hout, ook van zilver, staal of goud, Rembrandt-broches, Rembrandt-spelden. Om voor een groot genie te gelden, Rembrandts leven bij de vleet, naar ieders smaak gekneed.’ Het beeld was aan de noordzijde van de Botermarkt geplaatst bij het Reguliersplein (ofwel Kaasplein). In 1876 werd op dit plein het beeld van Thorbecke onthuld en veranderde de naam Reguliersplein in Thorbeckeplein. Rembrandt moest in verband met de komst van Thorbecke verhuizen naar het midden van de Botermarkt dat bij die gelegenheid omgedoopt werd in Rembrandtplein.
